抗體介紹：Tryptases comprise a family of trypsin-like serine proteases, the peptidase family S1. Tryptases are enzymatically active only as heparin-stabilized tetramers, and they are resistant to all known endogenous proteinase inhibitors. Several tryptase genes are clustered on chromosome 16p13.3. These genes are characterized by several distinct features. They have a highly conserved 3' UTR and contain tandem repeat sequences at the 5' flank and 3' UTR which are thought to play a role in regulation of the mRNA stability. These genes have an intron immediay upstream of the initiator Met codon, which separates the site of transcription initiation from protein coding sequence. This feature is characteristic of tryptases but is unusual in other genes. The alleles of this gene exhibit an unusual amount of sequence variation, such that the alleles were once thought to represent two separate genes, beta II and beta III. Beta tryptases appear to be the main isoenzymes expressed in mast cells, whereas in basophils, alpha-tryptases predominate. Tryptases have been implicated as mediators in the pathogenesis of asthma and other allergic and inflammatory disorders.

Anti-TPSB2抗體,肥大細(xì)胞類*β2抗體操作步驟
1．切片常規(guī)脫蠟至水。如需抗原修復(fù),可在此步后進(jìn)行
⒉ 緩沖液洗 3min/2 次。
⒊ 為了降低內(nèi)源性過(guò)氧化物酶造成的非特異性背景染色,將切片放在 Hydrogen Peroxide Block 中孵育 10-15 分鐘。
⒋ 緩沖液洗 5min/2 次。
⒌ 滴加 Ultra V Block , 在室溫下孵育 5 分鐘以封閉非特異性的背景染色。
（注：孵育不要超過(guò) 10 分鐘,否則會(huì)導(dǎo)致特異性染色降低。如果一抗的稀釋液中含有 5 － 10% 正常羊血清,這一步可以省略。）
⒍ 緩沖液洗 5min/2 次。
⒎ 滴加一抗工作液 37 ℃孵育 1 － 2 小時(shí)。（具體孵育時(shí)間和溫度由試驗(yàn)者zui終決定）
⒏ 緩沖液洗 5min/2 次。
9 ． 滴加 Primary Antibody Enhancer（增強(qiáng)子）,在室溫下孵育 20 分鐘。
10 ．緩沖液洗 5min/2 次。
11 ．滴加 HRP Polymer（酶標(biāo)二抗） ,在室溫下孵育 30 分鐘。
（注：HRP Polymer 對(duì)光敏感,應(yīng)避免不必要的光暴露并儲(chǔ)存在不透明的小瓶中。）
12 ．緩沖液洗 5min/2 次。
13 ．向 1ml DAB Plus Substrate （或 AEC Plus Substrate) 中滴加 1-2 滴 DAB Plus Chromogen （或 AEC Plus Chromogen）,混勻后滴加到切片上,孵育 3 － 15 分鐘。（具體時(shí)間由染色深淺決定。）
14 ．自來(lái)水充分沖洗,復(fù)染,脫水,透明,封片。
