NOGO相互作用線粒體蛋白1抗體,Anti-NIMP抗體背景介紹:Appears to be a potent inhibitor of regeneration following spinal cord injury.Nogo is an oligodendrocyte-specific member of the Reticulon family and is a component of CNS white matter that inhibits axon outgrowth, induces collapse of growth cones of chick dorsal root ganglion cells, and inhibits the spreading of 3T3 fibroblasts. Nogo is expressed by oligodendrocytes but not by Schwann cells, and associates primarily with the endoplasmic reticulum. Nogo exists in three different splice forms, Nogo-A, -B and -C. NIMP (NOGO-interacting mitochondrial protein), also known as RTN4IP1 (Reticulon-4-interacting protein 1), is a 396 amino acid mitochondrial protein that contains a C-terminal oxidoreductaselike domain and numerous sites for phosphorylation. NIMP is expressed in mitochondrial-rich tissue such as kidney, heart, skeletal muscle and specific regions within the nervous system. Through interaction with Nogo, it is likely that NIMP plays a role in Nogo-induced inhibition of neurite growth. There are three isoforms of NIMP that are produced as a result of alternative splicing events.

When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

一、抗原抗體反應(yīng)特點
1.高度特異性,特異性是抗原抗體反應(yīng)的zui主要特征,特定抗原決定簇只與特定抗體結(jié)合;
2.可逆性:抗原與抗體結(jié)合形成抗原抗體復(fù)合物的過程是一種動態(tài)平衡, 其反應(yīng)式為:Ag+AbAg·Ab;
3.適當濃度和比例;
4.敏感性:在測定血清中某一物質(zhì)的含量時,化學(xué)比色法的敏感度為mg/ml水平,酶反應(yīng)測定法的敏感度約為5~10μg/ml,免疫測定中凝膠擴散法的敏感度與酶反應(yīng)法相仿。
二、影響抗原抗體反應(yīng)的因素
1.電解質(zhì) :抗原抗體特異結(jié)合后,膠體粒便可發(fā)生脫水作用,此時易受電解質(zhì)影響,如有適當濃度的電解質(zhì)存在就會使它們失去部分負電荷而相互凝聚,于是出現(xiàn)明顯的凝集或沉淀現(xiàn)象。
2.酸堿度:合適的pH為6-8。
3.溫度:37℃通常是抗原體反應(yīng)zui適溫度。
免疫組化染色失敗的幾種原因
1.所染的全部切片均為陰性結(jié)果,包括陽性對照在內(nèi),全部呈陰性反應(yīng),原因可能是:(1)染色未按嚴格操作步驟進行;(2)漏加一種抗體,或抗體失效;(3)緩沖液內(nèi)含*,抑制了酶的活性;(4)底物中所加H2O2量少或失效;復(fù)染或脫水劑使用不當。
2.所有切片均呈弱陽性反應(yīng):(1)切片在染色過程中抗體過濃,或干燥了;(2)緩沖液配制中未加氯化鈉或pH不準確,洗滌不*;(3)使用已變色的呈色底物溶液,或呈色反應(yīng)時間過長;(4)抗體溫育的時間過長;(5)H2O2濃度過高,呈色速度過快;(6)粘附劑太厚。
3.所有切片背景過深:(1)未用酶消化處理切片;(2)切片或涂片過厚;(3)漂洗不夠;(4)底物呈色反應(yīng)過久;(5)蛋白質(zhì)封閉不夠或所用血清溶血;(6)使用全血清抗體稀釋不夠。
4.陽性對照染色良好,檢測的陽性標本呈陰性反應(yīng),固定和處理不當是zui常見的原因。對于陽性結(jié)果的定量判斷,常規(guī)方法是根據(jù)呈色深淺和陽性細胞數(shù)量分類計數(shù),以(-)、+、++、+++等分級和計數(shù)統(tǒng)計。
