腦源神經(jīng)營養(yǎng)因子抗體背景介紹:Neurotrophins function to regulate naturally occurring cell death of neurons during development. The prototype neurotrophin is nerve growth factor (NGF), originally discovered in the 1950s as a soluble peptide promoting the survival of, and neurite outgrowth from, sympathetic ganglia. More recently, three additional structurally homologous neurotrophic factors have been identified. These include brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT-3) and neurotrophin-4 (NT-4), also designated NT-5. These various neurotrophins stimulate the in vitro survival of distinct but partially overlapping populations of neurons. The Trk A receptor is the preferential receptor for NGF, but also binds NT-3 and NT-4. The Trk B receptor binds equally well to both BDNF and NT-4 and to a lesser extent NT-3, while the Trk C receptor only binds NT-3. BDNF promotes the survival of neuronal populations that are all located either in the central nervous system or directly connected to it. Belongs to the NGF-beta family.

1、染色過強(qiáng)?
原因:抗體濃度過高或孵育時間過長
解決方法:降低抗體滴度、抗體孵育時間:室溫1小時或4度過夜
原因:孵育溫度過高超過37度:
解決方法:一般在室溫20-28度
原因:DAB顯色時間過長或濃度過高?
解決方法:顯色時間不超過5-12分鐘,以顯微鏡下觀察為準(zhǔn)
2、非特異性背景染色?
原因:操作過程中沖洗不充分
解決方法:每步?jīng)_洗3次每次5分鐘
原因:組織中含過氧化物酶未阻斷
解決方法:可再配置新鮮3%H2O2封閉孵育時間延長
原因:組織中含內(nèi)原性*
解決方法:正常非免疫動物血清再封閉
原因:血清蛋白封閉不充分

3、染色弱?
原因:抗體濃度過低、孵育時間過短
解決方法:提高抗體濃度、孵育時間不能少于60分鐘
原因:試劑超過有效使用時間
解決方法:應(yīng)更換試劑
原因:操作中添加試劑時緩沖液未瀝干
解決方法:每步滴加試劑前瀝干切片中多余的緩沖液 使試劑稀釋 (應(yīng)防止切片干燥)
原因:室溫太低,低于15度
解決方法:要改放在37度孵育箱孵育30-60分鐘或4度冰箱過夜
原因:蛋白封閉過度
解決方法:封閉不要超過12分鐘
4、染色陰性?
原因:操作步驟錯誤
解決方法:應(yīng)重試,設(shè)陽性對照
原因:組織中無抗原
解決方法:設(shè)陽性對照以驗(yàn)證試驗(yàn)結(jié)果
原因:一抗與二抗種屬連接錯誤
解決方法:仔細(xì)確定一抗二抗種屬無誤
